    Different adaptation strategies of two citrus scion/rootstock combinations in response to drought stress

    Scion/rootstock interaction is important for plant development and for breeding programs. In this context, polyploid rootstocks presented several advantages, mainly in relation to biotic and abiotic stresses. Here we analyzed the response to drought of two different scion/rootstock combinations presenting different polyploidy: the diploid (2x) and autotetraploid (4x) Rangpur lime (Citrus limonia, Osbeck) rootstocks grafted with 2x Valencia Delta sweet orange (Citrus sinensis) scions, named V/2xRL and V/4xRL, respectively. Based on previous gene expression data, we developed an interactomic approach to identify proteins involved in V/2xRL and V/4xRL response to drought. A main interactomic network containing 3,830 nodes and 97,652 edges was built from V/2xRL and V/4xRL data. Exclusive proteins of the V/2xRL and V/4xRL networks (2,056 and 1,001, respectively), as well as common to both networks (773) were identified. Functional clusters were obtained and two models of drought stress response for the V/2xRL and V/4xRL genotypes were designed. Even if the V/2xRL plant implement some tolerance mechanisms, the global plant response to drought was rapid and quickly exhaustive resulting in a general tendency to dehydration avoidance, which presented some advantage in short and strong drought stress conditions, but which, in long terms, does not allow the plant survival. At the contrary, the V/4xRL plants presented a response which strong impacts on development but that present some advantages in case of prolonged drought. Finally, some specific proteins, which presented high centrality on interactomic analysis were identified as good candidates for subsequent functional analysis of citrus genes related to drought response, as well as be good markers of one or another physiological mechanism implemented by the plants.     Produtividade e características físico-químicas dos frutos da lima ácida 'Tahiti' sob diferentes disposições de microaspersores

    An adequate water management and irrigation system are important points to be considered for yield maximization and fruit quality of 'Tahiti' acid lemon in semi arid. The work focused in evaluation of the influence of different microsprinkler systems on yield and on physical chemical characteristics of fruits, in the different quadrants of the plant, of 'Tahiti' lemon acid orchard, with four years and planted at spacing of 5.0 x 7.0 m. The work followed a randomized block design with three treatments and six replications. Three arrangements of microsprinkler were: T1 - one emitter for each plant, located between plants along plant row; T2 - one emitter for two plants, located between plants along plant row; and T3 - one emitter for each plant, located at 0,3 m distant from the plant. The mean productivity values were signiï¬cantly different between the treatments T2 (19.506 kg ha ) and T1 (13.545 kg ha ). Fruits from T3 and T2 presented higher ascorbic acid contents than fruits from the treatment T1. The values of soluble solids, mean fruit weight and citric acid showed signiï¬cant differences among the quadrants of the plant, demonstrating the influence of the arrangement of the microsprinklers on the physical chemical parameters of the fruits of the 'Tahiti' acid lemon, under semi arid conditions of cultivation.O adequado manejo de água de irrigação é fundamental para maximização da produção e melhoria da qualidade dos frutos da lima ácida 'Tahiti' no semiárido. O trabalho visou avaliar a influência de diferentes disposições de microaspersores, na produtividade e características físico-químicas dos frutos, nos diferentes quadrantes da planta, num pomar de lima ácida 'Tahiti', com quatro anos, plantada no espaçamento de 5,0 x 7,0 m. O delineamento experimental foi em blocos casualizados, sendo: T1 – um microaspersor por planta, localizado entre plantas ao longo da fileira; T2 – um microaspersor para duas plantas, também ao longo da fileira; e T3 – um microaspersor por planta, localizado a 0,3 m da planta, e seis repetições. Foram consideradas parcelas subdivididas, nas quais se avaliou os frutos nos quadrantes norte, sul, leste e oeste da planta. Os valores de produtividade média apresentaram diferença significativa entre os tratamentos T2 (19.506 kg ha ) e T1 (13.545 kg ha ). Os frutos do T3 e T2 apresentaram teor de ácido ascórbico superior aos do T1. Os valores de sólidos solúveis, peso médio de frutos e ácido cítrico apresentaram diferenças significativas entre os quadrantes, demonstrando influência da disposição dos microaspersores sobre os parâmetros físico-químicos dos frutos da lima ácida 'Tahiti', em condições semiáridas de cultivo

    Recurrent water deficit causes epigenetic and hormonal changes in citrus plants

    The present study evaluated the physiological, molecular and hormonal parameters from scion/rootstock interaction of citrus plants during recurrent water deficit. Responses of the Valencia (VO) scion variety grafted on two rootstocks with different soil water extraction capacities, Rangpur Lime (RL) and Sunki Maravilha (SM), during three successive periods of water deficit: plants exposed to a single episode of water deficit (WD1) and plants exposed to two (WD2) and three (WD3) recurrent periods of WD were compared. The combinations VO/RL and VO/SM presented polymorphic alterations of epigenetic marks and hormonal (i.e. abscisic acid, auxins and salicylicacid) profiles, which were particularly prominent when VO/SM plantswere exposed toWD3 treatment. Upon successive drought events, the VO/SM combination presented acclimatization characteristics that enable higher tolerance to water deficit by increasing transpiration (E), stomatal conductance (g s ) and photosynthetic rate (A), which in turn may have facilitated the whole plant survival. Besides providing comprehensive data on the scion/rootstock interactions upon successive stress events, this study brings the first dataset suggesting that epigenetic alterations in citrus plants triggered by recurrent water deficit lead to improved drought tolerance in this crop species.     Pervasive gaps in Amazonian ecological research

    Biodiversity loss is one of the main challenges of our time,1,2 and attempts to address it require a clear un derstanding of how ecological communities respond to environmental change across time and space.3,4 While the increasing availability of global databases on ecological communities has advanced our knowledge of biodiversity sensitivity to environmental changes,5–7 vast areas of the tropics remain understudied.8–11 In the American tropics, Amazonia stands out as the world’s most diverse rainforest and the primary source of Neotropical biodiversity,12 but it remains among the least known forests in America and is often underrepre sented in biodiversity databases.13–15 To worsen this situation, human-induced modifications16,17 may elim inate pieces of the Amazon’s biodiversity puzzle before we can use them to understand how ecological com munities are responding. To increase generalization and applicability of biodiversity knowledge,18,19 it is thus crucial to reduce biases in ecological research, particularly in regions projected to face the most pronounced environmental changes. We integrate ecological community metadata of 7,694 sampling sites for multiple or ganism groups in a machine learning model framework to map the research probability across the Brazilian Amazonia, while identifying the region’s vulnerability to environmental change. 15%–18% of the most ne glected areas in ecological research are expected to experience severe climate or land use changes by 2050.     Rationale, study design, and analysis plan of the Alveolar Recruitment for ARDS Trial (ART): Study protocol for a randomized controlled trial

    Background: Acute respiratory distress syndrome (ARDS) is associated with high in-hospital mortality. Alveolar recruitment followed by ventilation at optimal titrated PEEP may reduce ventilator-induced lung injury and improve oxygenation in patients with ARDS, but the effects on mortality and other clinical outcomes remain unknown. This article reports the rationale, study design, and analysis plan of the Alveolar Recruitment for ARDS Trial (ART). Methods/Design: ART is a pragmatic, multicenter, randomized (concealed), controlled trial, which aims to determine if maximum stepwise alveolar recruitment associated with PEEP titration is able to increase 28-day survival in patients with ARDS compared to conventional treatment (ARDSNet strategy). We will enroll adult patients with ARDS of less than 72 h duration. The intervention group will receive an alveolar recruitment maneuver, with stepwise increases of PEEP achieving 45 cmH(2)O and peak pressure of 60 cmH2O, followed by ventilation with optimal PEEP titrated according to the static compliance of the respiratory system. In the control group, mechanical ventilation will follow a conventional protocol (ARDSNet). In both groups, we will use controlled volume mode with low tidal volumes (4 to 6 mL/kg of predicted body weight) and targeting plateau pressure <= 30 cmH2O. The primary outcome is 28-day survival, and the secondary outcomes are: length of ICU stay; length of hospital stay; pneumothorax requiring chest tube during first 7 days; barotrauma during first 7 days; mechanical ventilation-free days from days 1 to 28; ICU, in-hospital, and 6-month survival. ART is an event-guided trial planned to last until 520 events (deaths within 28 days) are observed. These events allow detection of a hazard ratio of 0.75, with 90% power and two-tailed type I error of 5%. All analysis will follow the intention-to-treat principle. Discussion: If the ART strategy with maximum recruitment and PEEP titration improves 28-day survival, this will represent a notable advance to the care of ARDS patients.
